Was ist die Idee der Hemmung der Cholesterinsynthese?
IJ: Mein Teil der VL beinhaltet die relevante Informationen nicht (absichtlich). Die Rolle des Insulins ist komplexer. Über Insig-1 scheint Insulin die Cholesterinsynthese zu hemmen, da Insig-1 den SCAP/SREBP Komplex stabilisiert und die Prozessierung («Reifung») im Golgi nicht stattfinden kann. Allerdings ist das Hauptelement der Regulation nicht Insig selbst sondern die Cholesterinkonzentration. Eine hohe Cholesterinkonzentration erhöht signifikant die Stabilität des unproduktiven Insig/SCAP/SREBP Komplex = SREBP-1 ist inaktiv.
Gleichzeitig treibt Insulin die Cholesterinsynthese an, und zwar durch Dephosphorylierung und Aktivierung der HMG-CoA-Reduktase (geschwindigkeitsbestimmendes Enzym). Hohes Cholesterin inaktiviert die HMG-CoA-Reduktase.
Sie sehen, die Effekte sin entgegengerichtet. «Der Sinn» ist vielleicht, dass hier «Sicherungen» «eingebaut» worden sind. Beides, zu viel Cholesterin und zu wenig Cholesterin, ist nicht gut.
